ZY GelStar Gel Extraction Kit

Rapid recovery of high-quality DNA fragments from 100 bp–9 kb

The specially treated silica membrane and efficient sol-gel system allow for simple and rapid recovery of high-quality DNA from TAE or TBE agarose gels. Each adsorption column can specifically bind up to 10 μg of DNA, without adsorbing enzymes, mineral oil, or other impurities.

The recovery rate for 100 bp–9 kb fragments is about 50%–80%, with a maximum exceeding 80%; the recovered DNA can be directly used for restriction digestion, ligation, transformation, PCR templates, sequencing, and library construction.

  • The entire purification process takes as little as 13–18 minutes.
  • All steps are performed at room temperature, with no ice bath required.
  • Special silica membranes show minimal batch-to-batch variation and good reproducibility.
Agarose Gel DNA Extraction

Direct purification of PCR products.

A dual-use box, purify PCR amplification products without cutting the gel

Can purify DNA fragments directly from the PCR amplification system. After adding 5 times the volume of binding buffer onto the column, high-purity products can be obtained through adsorption, washing, and elution.

Suitable for various DNA samples such as PCR products, plasmid restriction fragments, ligation products, and linearized plasmids. Purified products can be used immediately for downstream experiments or stored at -20°C.

  • Maximum adsorption capacity per column: 10 μg
  • Recommended elution: 30–50 μL
  • Compatible with routine enzymatic reactions and sequencing applications

Procedure

Supports both agarose gel DNA recovery and direct purification of PCR products.

Part 1 Agarose Gel DNA Recovery

Gel cutting, gel dissolving, binding, washing, and elution
  • Gel Dissolving
Cut the target band and weigh it; add 150 μL of binding buffer per 100 mg of 1% gel, 55°C until completely melted
  • Load onto column for binding
Transfer to the gel recovery column, stand at room temperature for 2 minutes, centrifuge at 8000 rpm for 1 minute
  • Wash to remove alcohol
Wash twice with 500 μL of wash solution; centrifuge the empty column at 12000 rpm and open the lid to evaporate ethanol
  • Elution and recovery
Add 30–50 μL of preheated elution buffer to the center of the membrane, incubate at 37°C for 2 minutes, then centrifuge at 12000 rpm for 1 minute

Part 2 PCR product purification

Binding, adsorption, washing, elution, and detection
  • Add binding buffer
Add PCR reaction solution to 5 times the volume of binding buffer and mix thoroughly
  • Load onto column for binding
Transfer to the gel recovery column, stand at room temperature for 2 minutes, centrifuge at 8000 rpm for 1 minute
  • Wash to remove alcohol
Wash twice with 500 μL washing solution; centrifuge the empty column and leave the lid open for 10 minutes
  • Elution detection
Add 30–50 μL preheated elution buffer, centrifuge after 2 minutes at 37°C; take 2–5 μL for electrophoresis detection

Storage, Shipping, and Scope of Application

Stable storage and transport, compatible with common DNA purification scenarios
  • Room temperature storage
Dry storage at 15–25℃ is stable for 1.5 years.
  • Long-term storage
When the ambient temperature is too high or for long-term storage, keep at 2–8℃.
  • Transportation Conditions
Supports room temperature transport; if precipitation occurs at low temperature, return to room temperature, and if necessary, dissolve with a 37℃ water bath.
  • Conventional fragments
Recovery rate of 100 bp–9 kb fragments is about 50%–80%.
  • Long fragments
10–15 kb recovery rate is about 20%–50%, it is recommended to increase the sample amount, elute with 50 μL and heat elute at 65°C.
  • Scope of Use
For research use only, not for clinical diagnosis, treatment, food or drugs.

Frequently Asked Questions

Common questions about recovery rate, binding, elution, and downstream reactions
What should I do if the gel recovery rate is low?
Check whether ethanol has been added to the wash solution; if the sol liquid turns orange-red or pink (pH > 7.5), add 10 μL of 3 M NaAc (pH 5.0) to adjust it back to light orange-yellow; it is recommended to use freshly prepared TAE/TBE. For long fragments, you can increase the loading volume, increase the elution volume to 50 μL, and use 65°C heating for elution.
What should be done if PCR product purification efficiency is low?
Add a binding solution equivalent to 5 times the volume of the PCR system and mix thoroughly; confirm that ethanol has been added to the wash solution. If the binding solution turns pink due to alkalinity, add 5 μL of 3 M NaAc (pH 5.2) to adjust it back to light orange-yellow. It is recommended to use Elution Buffer (2.5 mM Tris-HCl, pH 8.5).
What should be done if elution is poor or fails?
Use a low-salt elution buffer with a pH of 8.0–8.5, accurately add the elution buffer to the center of the adsorption membrane and ensure it is fully soaked; preheating the elution buffer to 65℃ before elution can further improve the results.
What should I do if there are still small fragments or primer residues after purification?
Fragments larger than 50 bp may not completely remove primer dimers during direct purification; if there are many nonspecific bands or the template is complex, please use the Part 1 agarose gel recovery procedure.
What if the downstream enzyme digestion or ligation of recovered DNA is poor?
A common reason is ethanol residue. After washing, be sure to centrifuge the empty column at 12,000 rpm for 1 minute and leave it open for 5–10 minutes to completely remove the alcohol. If the electrophoresis shows a smeared band, it may contain ssDNA; you can renature it by heating at 95℃ for 1 minute followed by slow cooling.
