Procedure

Using a 6-well plate as an example
  • Plating and Medium Change
Seed the day before; at the time of transfection, confluency should be 50%–70%; about 0.4×10⁶ 293T cells per well. Replace with 2 mL of fresh complete medium before transfection.
  • Dilute separately
Add 4 μg plasmid DNA to 250 μL DMEM; in another tube, add 10 μL reagent to 250 μL DMEM, mix gently, and let stand at room temperature for 5 min.
  • Prepare and add the complex
Mix equal volumes, mix gently, and incubate at room temperature for 20 min; add 500 μL DNA-liposome complex evenly to the cells and gently shake to mix.
  • Culture and detection
After 6–12 h, replace with fresh complete medium, continue culture for 24–40 h, and detect expression. The complex should not be vortexed or centrifuged; antibiotics are recommended to be removed for sensitive cells.

Frequently Asked Questions FAQ

Common Issues in DNA Transfection Experiments
What is the appropriate cell density before transfection?
For adherent cells, 50%–70% confluence is recommended; for suspension cells, a density of 2×10⁶–4×10⁶ cells/mL. Both overly high and overly low densities will reduce efficiency.
Is serum-free medium required during transfection?
It is recommended to use serum-free medium when diluting DNA and transfection reagents; adding serum-containing medium after the complex is formed usually does not affect efficiency.
Can antibiotics be added to the medium?
Antibiotics may reduce activity by increasing cell permeability, so it is recommended to avoid using them in the transfection system.
What should I do if transfection efficiency is low?
Check cell condition, plasmid purity (OD₂₆₀/OD₂₈₀ ≥ 1.8), and endotoxin levels, and optimize the DNA/reagent ratio and cell density.
What causes cell death after transfection?
Common reasons include poor cell condition, impure plasmid, high endotoxin levels, excessive reagent, or untimely medium change. It is recommended to adjust the amounts and replace with fresh medium promptly.
