ZY RulerStar Pre-stained DNA Marker

Covers the full range from 100 bp to 12 kb

ZY RulerStar Pre-stained DNA Marker is a pre-stained, ready-to-use DNA ladder launched by Zhi Yin Biologicals for molecular biology research, specifically designed for agarose gel electrophoresis. The product is composed of a mixture of independent linear double-stranded DNA fragments derived from digested plasmids, pre-mixed with 1× Loading Buffer, and embedded with GelRed nucleic acid dye.

Directly load 3–5 μL without additional staining; bands are clearly visible under a UV lamp, eliminating the need for post-staining with EB/SYBR and the issues of exposure shielding. The series includes four specifications, covering mainstream applications from 100 bp to 12 kb.

Procedure

Complete pre-stained DNA marker electrophoresis in four steps
  • Gel Preparation
Select the corresponding gel concentration according to the target fragment; no nucleic acid dyes need to be added.
  • Loading
Take the marker out from 4°C, let it equilibrate to room temperature, mix gently, and load 3–5 μL per lane directly.
  • Electrophoresis
Use 1×TAE or 0.5×TBE, 4–10 V/cm (7 V/cm recommended), until the front dye migrates to 2/3 of the gel.
  • Imaging
Directly place the gel under UV light to observe; the bands appear orange-red (GelRed channel), no post-staining required.

Frequently Asked Questions

DNA Marker usage and storage Q&A
Is ZY RulerStar DNA Marker compatible with GelRed / GelGreen / SYBR dyes?
Yes. The product is pre-stained with GelRed and is also compatible with GelGreen, SYBR Safe, and SYBR Green I. For post-staining, please follow the dye manufacturer's procedures; during pre-staining, it is recommended to avoid adding extra nucleic acid dye to the gel, as it may affect the migration of small fragments.
What is the recommended loading amount for each lane? What happens if too much is loaded?
Recommended 5 μL per lane; for wide wells, it can be increased to 8–10 μL. Too much can cause band smearing and residual sample in the well; too little will result in weak signals for small bands.
How long can it be stored at 4°C? What is the effect of repeated freeze-thaw?
Stable for ≥6 months at 4°C; long-term storage at –20°C can last up to 2 years. It is recommended to aliquot; repeated freeze-thaw more than 5 times may cause small fragment degradation and reduced intensity of the bright bands.
Can the supplied 5×Loading Buffer be replaced with a regular product?
No. The marker itself contains GelRed; if the sample requires a dye, please use the 5×Loading Buffer provided with the product (contains dye).
What is the function of the bright reference band?
The bright band has a concentration 2.5 times that of the other bands, serving both as a visual positioning anchor and semi-quantitative reference; when the sample brightness is close to that of the bright band, the DNA concentration can be roughly estimated as 50–125 ng/5 μL.
Why is 100 bp-II plus recommended to be used with 0.5× TBE + 1.5–2.0% gel?
The 100 bp range bands are dense, requiring a high-concentration gel to ensure resolution; TBE provides better resolution than TAE for small fragment separation.
Is it normal for precipitate or turbidity to appear before use?
After long-term storage at 4°C, a small amount of EDTA salt precipitation or GelRed micro-aggregation is normal. Re-dissolve in a 37°C water bath for 5 min and gently mix.
Why does the marker float out of the loading well?
Common reasons include insufficient glycerol ratio, excessive use of buffer leading to decreased ionic strength, or excessive sample volume and insufficient density.
Can Marker be used on PAGE gels?
100 bp-II plus is recommended for 5% PAGE gels + 0.5×TBE; the other three types are mainly for agarose gels and are not recommended for PAGE.
