ZY GelRed Prestained Loading Buffer (5×)

Stain as you load; this process does not require additional EB, which helps reduce EB use and related contamination risks.

ZY GelRed Prestained Loading Buffer (5×) is a pre-stained loading buffer containing nucleic acid dye launched by Zhisheng Bio's ZY RulerStar series, specially designed for agarose gel nucleic acid electrophoresis. The product is optimized based on GelRed, combining nucleic acid staining with sample loading, allowing samples to be loaded directly after mixing.

After electrophoresis, the gel can be observed and photographed under UV light or blue light, without the need to add EB or other nucleic acid dyes to the gel. The buffer uses bromophenol blue and patented Blue V as indicators, and after dilution to 1× working concentration, it still maintains a relatively high density, allowing easy sample sinking, clear color, and convenient monitoring of electrophoresis progress.

Scope of Application and Downstream Uses

Covers routine nucleic acid electrophoresis, fragment analysis, and molecular cloning procedures
  • PCR product identification
Used for assessing the size of amplified fragments.
  • Plasmid DNA electrophoresis
Suitable for analyzing enzymatic digestion products and observing supercoiled, relaxed, or linear conformations.
  • Genomic DNA analysis
Used to observe integrity and degradation status.
  • Nucleic acid gel extraction
Used as a tracking indicator during the gel extraction process.
  • Routine molecular cloning
Applicable for library construction, restriction fragment selection, and other processes.

Instructions for Use

Mix evenly at a 4:1 ratio, load directly, and complete imaging
  • Prepare the sample
Take an appropriate amount of DNA sample, such as 4 μL, and add it to a centrifuge tube.
  • Mixed Buffer Solution
Add this product to the sample at a ratio of sample:buffer = 4:1, and mix gently by pipetting.
  • Load the sample directly
Pipette the mixture into agarose gel wells and perform electrophoresis.
  • Observe and photograph
After electrophoresis, observe and photograph under a UV lamp or compatible blue light transilluminator.
Sample loading amount prompt:Each 1 μL of DNA loading buffer can carry up to 500 ng of nucleic acid sample; exceeding this amount may cause significant changes in migration.

Tracking Dye Migration Reference

Equivalent DNA fragment length at different agarose concentrations under 0.5×TBE conditions
In 0.5×TBE electrophoresis buffer, the migration rate of bromophenol blue is approximately 2.2 times that of xylene cyanol and is independent of agarose concentration.
Agarose ConcentrationBromophenol BlueXylene Cyanol
0.6%1 kb/
1.0%0.6 kb2 kb
1.4%0.2 kb1.6 kb
2.0%0.15 kb/
In 0.4–1.5% agarose gels, bromophenol blue migrates at approximately the same rate as 300 bp double-stranded linear DNA, while xylene cyanol migrates at approximately the same rate as 4 kb double-stranded linear DNA. It is recommended to stop electrophoresis when the tracking dyes are about 1–2 cm from the bottom of the gel.

Precautions

Usage guidance, migration interpretation, safe operation, and storage recommendations
  • Matching Marker
It is recommended to use with verified compatible pre-stained DNA Markers; no EB or other nucleic acid dyes are required throughout.
  • Compatibility Optimization
When mixed with ordinary or other brand DNA Markers, necessary optimization is required to avoid migration differences affecting interpretation.
  • Migration Reference
The migration rate of bromophenol blue in 0.5× TBE is approximately 2.2 times that of xylene cyanol.
  • For research use
For scientific research use by professionals only. Not for clinical diagnosis, treatment, food, or drug use.
  • Storage conditions
For long-term storage, it is recommended to keep at -20°C; if precipitation occurs, equilibrate briefly at room temperature and mix before use.

Frequently Asked Questions

Common questions about staining safety, proportions, sample loading amounts, and migration positions
Do I still need to stain with EB or other nucleic acid dyes after electrophoresis?
No. This product already contains GelRed nucleic acid dye. Samples are stained upon loading and can be directly observed and photographed under UV or blue light after electrophoresis.
Is GelRed Prestained Loading Buffer safe? How is it different from EB?
GelRed has significantly lower mutagenicity than ethidium bromide and is safer. This product enables staining during loading through premixed GelRed, avoiding the risks of contact and waste treatment associated with EB.
What is the difference between 5× and the common 6× Loading Buffer?
This product is 5×, recommended sample:buffer ratio = 4:1; the common 6× is usually 5 μL sample plus 1 μL buffer. The ratio should be followed according to this product's instructions to avoid concentration deviations affecting migration.
How much loading buffer should be added? How much sample does 1 μL accommodate?
Recommended sample:buffer ratio = 4:1. Each 1 μL of loading buffer can hold up to 500 ng of nucleic acid sample; exceeding this will significantly affect migration.
Where is it appropriate for bromophenol blue and xylene cyanol to run?
In 0.5× TBE with 0.4–1.5% agarose gel, bromophenol blue corresponds approximately to 300 bp double-stranded DNA, and xylene cyanol corresponds approximately to 4 kb double-stranded DNA; it is recommended to stop electrophoresis when the tracking dyes are about 1–2 cm from the bottom of the gel.
